I’m going to take a wild guess and say your COR and/or CTCSS (and possible the duplex setting) is wrong.
Make sure duplex=0 or duplex=1 (see http://ohnosec.org/drupal/node/84).
Then at the Asterisk CLI do a "rpt stats <your node number>" and make sure the “Signal on input………..: NO/YES” is following your keyups.
