A couple of thoughts:

- Make sure you set the correct duplex mode, should be half duplex. duplex=0 or duplex=1, http://ohnosec.org/drupal/node/84 The URI red PTT light should not come on when you TX with your HT. 

- Are you on the right gm300 pins? I think COR, CTCSS and audio are selectable in the programing software. Start simple, have no CTCSS to start with then add it once you have COR working. 

- Look at the Asterisk CLI to see if you have “Signal On Input” when you key your HT.  *CLI>rpt stats <your node number> 

- What are your settings in simpleusb.conf or usbradio.conf?
