Im not sure exactly what your explaining so if you havent already done so, try changing ctcssfrom=dsp to ctcssfrom=no. I thought the blinking light on the URI was the PC heartbeat. Check the CLI screen when its slow to make sure asterisk starts correctly. Also check the sound driver.
